I knew of and played games of quite a few of the unlockeable characters. But others, like flywrench and Jill piqued my interest so I read up on them. I hope they might implement a few more in future updates that have unique traits, but don't break levels.

Pretty much I only knew of Alien Hominid, Commander Video, Josef, and of course Mr.Minecraft.

Only characters I really knew of before Meat boy were the Ninja, Pink Night, Head crab, Captain Viridian, Ogmo, Steve, and the Kid. The others I had never seen before.

I knew about them all except for Jill and Flywrench, but they were announced pretty early, so I was able to play their games before Meat Boy hit.  Also Aquaria very well be the greatest indie game of all time

I gotta say I am new to the indy game community and didn't know a lot of them.
But following the Let's Plays of "Raocow" at least let me know of Runman and the Captain

But yeah, everone I didn't know of I checked and played most of the games. I am actually going to buy machinarium, Josephs game.
A lot of great games got their indy characters in Meat Boy and I still have to play a few of these!
