I did Brimstone (CH4 Light) before Blood Clot (CH2 Dark) but probably found Blood Clot easier than Brimstone in the end. Hell has quite a few meticulous jumps that aren't super hard but certainly require some precision, and Dark Hospital just felt so much easier than I remembered when I revisited it. Of course, I like to use different characters, whomever I feel gives me the safest chance of beating a level, and if you're doing Meat Boy only, perhaps that would change things.
Dr. Fetus Boy (CH6 Dark) is probably harder than either of these two, it's only 5 levels, but I certainly found it a lot harder than its Light counterpart, you can find a lot of ways to die in those Dark levels (3/5 of them all take place above pits which can really add to your stress factor), and having to do No Quarter (6-5X) again every time you mess up gets annoying fast. Still it's not that bad, and with a little luck you can be through it quickly due to it only being 5 levels.

Actually, I didn't find Hell that hard at all. Well... not in a metaphorical sense, but even for the levels that were the most annoying for me, at least they were fun to play at the same time.  But yeah, it would probably be pretty hard Iron Boy running Hell. Dark Hospital, on the other hand, wasn't as bad. Plus, it had a lot of levels that could be broken with other characters, most of which are pretty obvious. I'd probably try that first, if I actually had Squirrel Boy.  Also, I took a look at your profile, and it seems you got Maggot Boy yesterday night! Congrats to you.

Got Brimstone Boy today, definitely the toughest yet. Level order was 15-20, 7-14, 1-6. Oddly enough considering how basic it is, 15 really did give me the most trouble. Lost a bunch of runs in 17, 18, 20, 7, and 4 as well. 4's definitely the oddball there as it really isn't that tricky, but I attribute it to nerves.
Think I'll go for Blood Clot next.

My difficulty level order is as such:
Wood Needle Salt Dead Squirrel Brimstone Dr Fetus Blood Clot Maggot Missile Demon
That's as far as I've gotten, but I'm assuming the final 3 will look like this:

I really wanna get Dr. Fetus Boy but I keep messing up at stupid things. I also wanna get Missile Boy, mainly cuz I want revenge against Ch 3 Dark World. Damn, I hate that place!
That would leave me with: Demon Boy Zombie Boy Girl Boy Impossible Boy
Out of these 4, which one is the easiest?

Well then, I got the Kid a little while ago, and ever since then, I've started to get more confident in my No-Death Runs. I just got Squirrel Boy today. Here is my order: 6-20, 1-5 I used The Kid for most of the run. Meat Boy was used only for one stage, 1-6X. After that, it was either the Kid, Viridan, or Naija. Naija got used for 1-18X, Viridian for 1-10X, and 1-14X. There were a couple others he (  ) could be used for, but I found them unreliable and only saved them for Speed Runs.  KEY:  = I got it!  = I got it, but it gave me some trouble.  = This is one I'm working on. Right Now.  = Maybe some day...  = Heh. You know the drill by know. Forest / Hospital / Factory / Hell / Rapture / The End / Cotton AlleyLight World:  /  /  /  /  /  / Dark World:  /  /  /  /  /  / I'll probably go for Blood Clot Boy next, considering Dark World Hospital didn't give me that much trouble the last time I tried it. After that, will be Brimstone Boy, then Dr. Fetus Boy. Hopefully by then, the level editor will be out!

Started practicing through the levels for Demon Boy and it feels like 80% of the difficulty is saturated in around 5 to 7 levels. I mean honestly, how can you look at Starless and say "Yeah, that's a fair level."? Demon maws spitting out like a machine gun, it's ridiculous. The fact that it's silhouetted makes it even worse because if I try to use Runman I often mistake him for a mini-maw. Also, Old Scratch, Thistle and Imperial can piss off.
I feel like for me this is going to be harder than Missile Boy because of my lack of drive to get it done. I hate the world, I hate the enemies, and I hate the music (I'm sure to catch hell for that, no pun intended).

Hit the halfway point with these today, getting my seventh Iron Boy with Maggot Boy. Order I ended up with was 9-20, 1-8, though I was doing 16-20, 9-15, 1-8 for a while as I found I couldn't not try for the shortcut in 5-16 and failure on that is death, but determined those stupid little zombie things were the biggest hurdle and wanted them out of the way as soon as possible. This one was frustrating as all the levels seemed very doable, but I kept dying in stupid ways. Lost a few late, included one on the very last sawblade before Bandage Girl in Descent. That one had me raging.
Will likely do more, Blood Clot's on deck probably, maybe Dr. Fetus.

I did like... the exact same thing you are describing here today myself, and Starless was definitely one of the levels that I was like, well, this is going to suck. Easy to finish once for completion, hard to do 100% reliably on a no-death run. I guess we'll see how it goes long term after some practice but with only Missile, Demon, Zombie, or the Cotton Alley runs left to choose from, these runs are getting pretty ruthless.

That run was made using Meat Ninja and as far as I know, he cannot be used to complete no death runs. I assume Steve is also treated that way. Also, I feel it's a way too hard drop to be made with a legit character during a no death run.

Not if you're only using Meat Boy. Demon Boy is only easy because Viridian breaks half the levels. Missile Boy was a cake walk in comparison.

Shame, might have been useful. I hadn't noticed it was Meat Ninja, although the drop is possible with Meat Boy, since thats how I did it.

Yeah, it would help a little if it was easier to pull out. Meat Boy can do that too, yes, but only from the right side of the laser eye. I think the player passed that eye from the left side in the video.
